Chesky's announcement comes in the wake of the Halloween night shooting in Orinda, California, on Thursday that left five people dead.
He further tweeted what steps will be taken by the San Francisco-based company: "First, we are expanding manual screening of high-risk reservations flagged by our risk detection technology".
Chesky said Airbnb is also forming a rapid response team dedicated to house parties.
Users who violate the updated guest policies will face immediate action and removal.
Chesky said he has appointed Margaret Richardson from the Airbnb executive team to oversee the new team and initiative a 10-day sprint to develop and implement the new safety initiatives.
The company will remove guests who fail to comply, Chesky said.
Writing on Twitter on Saturday, Mr. Chesky said: "What happened on Thursday night in Orinda, CA bad I feel for the family and neighbors affected by this tragedy - we are working to support them".

The woman who rented the home lied to her Airbnb host, saying she was renting it so her asthmatic family members could escape wildfire smoke, according to a person with knowledge of the transaction, who was not authorized to publicly disclose the information and spoke only on condition of anonymity.

Three people, all from the Bay Area, died at the scene and a fourth died at the hospital, authorities initially said.
The sheriff's department said they were responding to a noise complaint at the house around the time the shooting was reported. Two people died at the hospital.
Mr Wang's wife then got in touch with the renter, who said there were only a dozen people at the home, which was clearly not the case as the owners could see more people than that through a video doorbell fitted at the home.
The party was advertised on social media as an "Airbnb mansion party".
"We called the police", he told the paper.
