The suspect, James Michael Bowden, 32, took shelter in the home, about 5 miles southwest of the small city of Dardanelle after the deputy was shot, police said.
Bowden had been barricaded inside a residence on a rural road in Yell County with a woman who may have been a hostage, Arkansas Police spokesman Bill Sadler told a news conference.
Bowden released the hostage about 30 minutes before he gave himself up to law enforcement.
Arkansas Attorney General Leslie Rutledge also responded to Mainhart's death in a statement, referencing the deputy's mission to his community.
Arkansas State Police identified the deceased officer as Lt. Kevin C. Mainhart, 46, of the Yell County Sheriff's Department. Authorities allege that Bowden shot and killed the deputy during the traffic stop.
Lt. Kevin Mainhart, who was serving as a sheriff's deputy, has been with West Memphis police for 20 years, before moving back home to the River Valley.

Sadler said authorities believe the traffic stop and the disturbance call are connected. Sadler said Bowden let one female hostage go during the standoff and she is being questioned. He had previously worked for the West Memphis Police Department for more than twenty years, according to a State Police news release.
After the suspect was arrested, he was taken to a hospital and is expected to be transported to the Conway County jail, authorities said.
After police found Mainhart's body, they found the bodies of two unidentified women at a nearby home where the disturbance had been reported. A previous story was unclear on whether the bodies were found inside or outside. The incident happened at the intersection of State Highway 27 and Slow Fork Road near Chickalah in Yell County.
The fatal shooting of Mainhart was on the same day of the 36th annual memorial to honor fallen officers at the Arkansas state Capitol.
During the course of the traffic stop, Bowden, who was driving the vehicle, critically shot Mainhart, then drove away, leaving him for dead.
