The news comes a year after former co-host Michael Strahanleft the show after four years to take on a full-time role on ABC's Good Morning America.
"Live with Kelly and Ryan" airs weekdays at 9 a.m. on WEWS Channel 5.
Megyn Kelly and Kelly Ripa are set to go head-to-head over the mid-morning TV audience starting in the fall and perhaps steal some thunder from the traditional morning shows on earlier.
"It's good timing for me because it's my 21st wedding anniversary, the same day we are announcing our new co-host", Ripa said. The host of everything is already slated to host Dick Clark's New Year's Rockin' Eve this year once again.
Seacrest will also continue his nationally syndicated radio shows for iHeartMedia, broadcasting from ABC in NY.

Ryan Seacrest is joining Disney-ABC's syndicated morning show "Live" as Kelly Ripa's co-host.
When they finally came onstage together, Seacrest was all smiles sitting in his new co-host chair.
Live is based in New York City. Seacrest will now record his Top 40 radio show inside the building where Live with Kelly and Ryan films.
"I would first like to thank all of the men and women who sat next to me cohosting with me this past year", Ripa said.
